The number of different bacterial species can only be roughly estimated, but millions of them probably exist on earth. Many are essential for life, but some are extremely dangerous, e.g., foodborne pathogens such as Salmonella spp. or Listeria monocytogenes.

No microbiological food testing is complete without these two species.

  • DNA extraction from Listeria

Applicability

  • Fast and straightforward extraction of PCR-grade genomic DNA from Listeria cultures.
  • Tested with the Listeria monocytogenes with 139E variant qPCR detection kit from MAMBA Diagnostik GmbH.
  • Shelf life: at least one year at 20 °C
  • DNA extraction from cultured cells culture in under than 30 minutes.
  • Extracted DNA remains stable for at least one week when stores at room temperature, briefly vortex 3-5 sec. before use.
  • Only three pipetting steps required to extract DNA, minimizing hands-on time and significantly reducing the risk of contamination.
  • Requires basic laboratory equipment: centrifuge, vortex mixer and standard pipettes.

Specificity

Optimized specifically for Listeria monocytogenes. The extraction procedure allows effective cell lysis, extraction, and purification of DNA.

Sensitivity

Clear and reliable detection of Listeria is achieved even when the culture has grown to only 10%. No difference in signal intensity is observed when using cultures grown to 50%.

Sensitivity results

Multiple culture yields were tested with qPCR:

  • 100% = 1000 µl culture
  • 50% = 500 µl culture + 500 µl cell free medium
  • 10% = 100 µl culture + 900 µl cell free medium
